Analytical confirmation usually relies on reversed-phase high-performance liquid chromatography for purity and on liquid chromatography coupled to mass spectrometry for identity. Mass data reveal the expected molecular mass and can flag truncated or oxidized species. Amino acid analysis and peptide mapping provide sequence-level verification. Immunoassays are used in some biological matrices, but antibodies raised against one releasing-hormone analog may cross-react with another. Reported purity figures depend heavily on the method used, so comparisons between suppliers require matching the analytical approach.
The two variants differ dramatically in how long they persist in circulation. The form lacking the albumin-binding group has a plasma half-life measured in tens of minutes, comparable to the natural hormone fragment. The version carrying the drug affinity complex binds albumin and shows a half-life of roughly six to eight days in human studies. That figure comes from small trials that tracked hormone levels over extended periods. The physiological consequences of sustained versus pulsatile stimulation are still debated and the literature does not settle the point.
Lyophilized peptide powder is comparatively stable when kept dry, cold, and protected from light. Once dissolved, the molecule is vulnerable to deamidation, oxidation, and aggregation, with the rate depending on pH, buffer composition, and temperature. Alkaline conditions and repeated freeze-thaw cycles accelerate loss of the intact peptide. The methionine present in the native sequence is a known oxidation site, which is one reason it was replaced in the modified fragment. Suppliers typically recommend cold storage of solutions and use within a short window.
CJC-1295 belongs to a family of synthetic peptides modeled on growth hormone-releasing hormone, the hypothalamic signal that prompts the pituitary to release growth hormone. The compound is built from the first twenty-nine amino acids of the natural human sequence, a fragment that retains full receptor binding capacity. Native growth hormone-releasing hormone is degraded quickly in circulation, so the fragment alone has limited practical value. Early work therefore focused on chemical modifications that preserve receptor binding while slowing enzymatic breakdown. The result is a molecule described in the literature as a long-acting analog of the natural hormone.
| Property | Value | Notes |
|---|---|---|
| Molecular mass | Approximately 3.4 to 3.6 kDa | Depends on whether the affinity complex is attached |
| Appearance | White to off-white lyophilized powder | Freeze-dried solid, often in a sealed vial |
| Solubility | Soluble in water and aqueous buffers | Dissolution rate varies with pH and buffer salt |
| Typical storage | Below minus 20 degrees Celsius, dry and dark | Dissolved material is usually kept cold and used promptly |
| Common analytical methods | Reversed-phase HPLC and mass spectrometry | Peptide mapping and amino acid analysis add sequence detail |
Reconstitution is typically performed with sterile water or bacteriostatic water, added slowly against the vial wall. The resulting solution should be clear and colorless; cloudiness or visible particles suggest a problem with the material or the diluent. Once in solution, the peptide is less stable than the dry powder. Refrigerated storage at two to eight degrees Celsius is common for short-term holding, while freezing aliquots is described for longer periods.
Purity is most often assessed by reversed-phase high-performance liquid chromatography, reported as a percentage of total peak area. Identity is confirmed by mass spectrometry, which yields a molecular ion consistent with the expected sequence. Amino acid analysis and peptide mapping provide additional characterization. Reported purity values are method-dependent, so figures from different laboratories are not always directly comparable without details of column, gradient, and detection wavelength.
Peptide degradation proceeds mainly through hydrolysis, oxidation of methionine, and deamidation of asparagine or glutamine residues. The maleimide group on the albumin-binding variant can also react with thiols or hydrolyze in aqueous media. Because these pathways accelerate with temperature and pH extremes, handling conditions strongly influence measured stability. Stability data in the public literature are limited and often generated under differing conditions, so general statements about shelf life should be read as approximate.
Binding to GHRH receptors on pituitary somatotroph cells triggers cyclic AMP signaling and stimulates growth hormone synthesis and release. Because the peptide acts upstream of the pituitary, effects are mediated through endogenous growth hormone rather than direct receptor activation in peripheral tissues. Increases in insulin-like growth factor 1 are generally described as a downstream consequence. Most published human exposure data come from small early-stage studies, and the clinical significance of the pharmacokinetic profile remains incompletely characterized.
CJC-1295 is a synthetic analogue of growth hormone-releasing hormone, built on the 29-amino-acid GHRH(1-29) fragment. Four substitutions distinguish it from the natural sequence: D-alanine at position 2, glutamine at position 8, alanine at position 15, and leucine at position 27. These changes reduce enzymatic cleavage and extend the peptide's persistence in circulation. The compound is discussed in two forms, one carrying a drug affinity complex and one without it.
The drug affinity complex is a maleimidopropionic acid group attached to a lysine side chain. It reacts with the free thiol of cysteine-34 on circulating albumin, forming a covalent bond. This conjugation keeps the peptide in the bloodstream and shields it from rapid renal filtration and proteolysis. Reported circulation half-lives for the albumin-bound form fall in the range of roughly six to nine days in early human studies.

Since thaumatin crystallizes very quickly and easily in the presence of tartrate ions, thaumatin-tartrate mixtures are frequently used as model systems to study protein crystallization. The solubility of thaumatin, its crystal habit, and mechanism of crystal formation are dependent upon the chirality of precipitant used. When crystallized with L- tartrate, thaumatin forms bipyramidal crystals and displays a solubility that increases with temperature; with D- and meso-tartrate, it forms stubby and prismatic crystals and displays a solubility that decreases with temperature. This suggests control of precipitant chirality may be an important factor in protein crystallization in general.
=== Fluorescence spectroscopy === Fluorescence spectroscopy is a highly sensitive method for studying the folding state of proteins. Three amino acids, phenylalanine (Phe), tyrosine (Tyr) and tryptophan (Trp), have intrinsic fluorescence properties, but only Tyr and Trp are used experimentally because their quantum yields are high enough to give good fluorescence signals. Both Trp and Tyr are excited by a wavelength of 280 nm, whereas only Trp is excited by a wavelength of 295 nm. Because of their aromatic character, Trp and Tyr residues are often found fully or partially buried in the hydrophobic core of proteins, at the interface between two protein domains, or at the interface between subunits of oligomeric proteins. In this apolar environment, they have high quantum yields and therefore high fluorescence intensities. Upon disruption of the protein's tertiary or quaternary structure, these side chains become more exposed to the hydrophilic environment of the solvent, and their quantum yields decrease, leading to low fluorescence intensities. For Trp residues, the wavelength of their maximal fluorescence emission also depend on their environment. Fluorescence spectroscopy can be used to characterize the equilibrium unfolding of proteins by measuring the variation in the intensity of fluorescence emission or in the wavelength of maximal emission as functions of a denaturant value. The denaturant can be a chemical molecule (urea, guanidinium hydrochloride), temperature, pH, pressure, etc. The equilibrium between the different but discrete protein states, i.e.
In Singapore, 7-Eleven forms the largest chain of convenience stores island-wide. There are 393 7-Eleven stores in the country as of February 2018. Stores in Singapore are operated by DFI Retail Group (formerly Dairy Farm International Holdings), franchised under a licensing agreement with 7-Eleven Incorporated. The first 7-Eleven store in Singapore was opened along Upper Changi Road in June 1983, and in 1986 the first franchised 7-Eleven store (under the Jardines) was opened. The license was then acquired by Cold Storage Singapore, a subsidiary of the Dairy Farm Group, in 1989. In 2006, Shell Singapore and 7-Eleven agreed to rebrand all 68 of its Shell Select convenience stores into 7-Eleven. The partnership was terminated in October 2017, and the remaining 52 7-Eleven stores in Shell petrol stations were gradually rebranded back into Shell Select.
Anatoly Lunacharsky became the first People's Commissar for Education of Soviet Russia. In the beginning, the Soviet authorities placed great emphasis on the elimination of illiteracy. All left-handed children were forced to write with their right hand in the Soviet school system. Literate people were automatically hired as teachers. For a short period, quality was sacrificed for quantity. By 1940, Stalin could announce that illiteracy had been eliminated. Throughout the 1930s, social mobility rose sharply, which has been attributed to reforms in education. In the aftermath of World War II, the country's educational system expanded dramatically, which had a tremendous effect. In the 1960s, nearly all children had access to education, the only exception being those living in remote areas. Nikita Khrushchev tried to make education more accessible, making it clear to children that education was closely linked to the needs of society. Education also became important in giving rise to the New Man. Citizens directly entering the workforce had the constitutional right to a job and to free vocational training. The education system was highly centralized and universally accessible to all citizens, with affirmative action for applicants from nations associated with cultural backwardness. However, as part of a general antisemitic policy, an unofficial Jewish quota was applied in the leading institutions of higher education by subjecting Jewish applicants to harsher entrance examinations.

Necrophages (also known as carrion feeders) are animals that feed on decomposing dead animal biomass, such as the muscle and soft tissue of carcasses or corpses. The term derives from Greek nekros, meaning 'dead', and phagein, meaning 'to eat'. Many hundreds of necrophagous species have been identified including invertebrates in the insect, malacostracan and gastropod classes and vertebrates such as vultures, hyenas, quolls and wolves. Necrophagous insects are important in forensic science as the presence of some species (e.g. Calliphora vomitoria) in a body, coupled with information on their development stage (e.g. egg, larva, pupa), can yield information on time of death. Information on the insect species present can also be used as evidence that a body has been moved, and analysis of insect tissue can be used as evidence that drugs or other substances were in the body. Necrophages are useful for other purposes too. In healthcare, green bottle fly larvae are sometimes used to remove necrotic (dead) tissue from non-healing wounds, and in waste management, black soldier fly larvae are used to convert decomposing organic waste into animal feed. Biotechnological applications for necrophage-derived genes, molecules and microbes are also being explored.

SUMOplot - online free access software developed to predict the probability for the SUMO consensus sequence (SUMO-CS) to be engaged in SUMO attachment. The SUMOplot score system is based on two criteria: 1) direct amino acid match to the SUMO-CS observed and shown to bind Ubc9, and 2) substitution of the consensus amino acid residues with amino acid residues exhibiting similar hydrophobicity. SUMOplot has been used in the past to predict Ubc9 dependent sites. seeSUMO - uses random forests and support vector machines trained on the data collected from the literature SUMOsp - uses PSSM to score potential SUMOylation peptide sites. It can predict sites followed the ψKXE motif and unusual SUMOylation sites contained other non-canonical motifs. JASSA - online free access predictor of SUMOylation sites (classical and inverted consensus) and SIMs (SUMO interacting motif). JASSA uses a scoring system based on a Position Frequency Matrix derived from the alignment of experimental SUMOylation sites or SIMs. Novel features were implemented towards a better evaluation of the prediction, including identification of database hits matching the query sequence and representation of candidate sites within the secondary structural elements and/or the 3D fold of the protein of interest, retrievable from deposited PDB files.
The hydrophobic effect represents the tendency of water to exclude non-polar molecules. The effect originates from the disruption of highly dynamic hydrogen bonds between molecules of liquid water. Polar chemical groups, such as OH group in methanol do not cause the hydrophobic effect. However, a pure hydrocarbon molecule, for example hexane, cannot accept or donate hydrogen bonds to water. Introduction of hexane into water causes disruption of the hydrogen bonding network between water molecules. The hydrogen bonds are partially reconstructed by building a water "cage" around the hexane molecule, similar to that in clathrate hydrates formed at lower temperatures. The mobility of water molecules in the "cage" (or solvation shell) is strongly restricted. This leads to significant losses in translational and rotational entropy of water molecules and makes the process unfavorable in terms of free energy of the system. In terms of thermodynamics, the hydrophobic effect is the free energy change of water surrounding a solute. A positive free energy change of the surrounding solvent indicates hydrophobicity, whereas a negative free energy change implies hydrophilicity. In this way, the hydrophobic effect not only can be localized but also decomposed into enthalpic and entropic contributions.
Experts estimated that some 303 billion barrels of oil, or about 17% of the world's reserves, were located in Venezuela. Much of it is ultra-heavy crude, a dense, viscous, and high-sulfur type likened to a "semi-solid tar". Many US refineries along the Gulf Coast process this type of crude oil from Canada and Mexico to make diesel, jet fuel, asphalt, and elements of petrochemicals. Venezuela's oil industry had been in decline since Hugo Chávez's wave of expropriations, during which former oil contracts were shifted to joint ventures with the state-owned Petróleos de Venezuela S.A. (PDVSA) and new fiscal rules were imposed. Legal instability and other risks drove away investors, which Venezuela could not compensate for because of its overreliance on PDVSA's "inefficient operations, economic instability, discretionary policies, and poor investor protection", worsened by US sanctions on the company since 2017. In this time, the industry's oil infrastructure declined, and China increased cheap imports from Venezuela. Analysts, experts, and oil industry officials believed reconstruction could cost billions of dollars and take two to seven years or more, requiring risky investment and a lengthy process of creating contracts. Experts also said that US companies' refining capability could threaten China's presence in the Venezuelan oil industry.

One femtomole is exactly 602214076 molecules; attomole and smaller quantities do not correspond to a whole number of entities. The yoctomole, equal to around 0.6 of an individual molecule, did make appearances in scientific journals in the year the yocto- prefix was officially implemented.
In accordance with such findings, high-dose CPA shows equivalent effects on the prostate gland in men relative to high-dose diethylstilbestrol or buserelin, which both achieve castrate levels of testosterone. However, a lower dosage of 50 mg/day CPA has been found to produce a reduction in prostate volume in men with benign prostatic hyperplasia that is reportedly comparable to that observed with surgical or medical castration. In accordance, the dosage of CPA that achieves complete inhibition of the secretory function of the healthy prostate gland is around 50 to 100 mg/day, which is less than the dosage of 200 to 300 mg/day CPA that is used to treat prostate cancer. It has been said that in combined androgen blockade regimens with castration and CPA as the AR antagonist for prostate cancer, due to the marked reduction in androgen levels, lower dosages of CPA than those used as a monotherapy would seem to be equally effective. Relative to the 200 to 300 mg/day dosage of CPA used as a monotherapy in prostate cancer, the recommended dosage in combined androgen blockade is 100 to 200 mg/day. It has been stated that this dosage should be more than necessary to inhibit the effects of the remaining adrenal androgens in castrated men. Despite considerable suppression of testosterone levels, only a modest suppression of spermatogenesis typically occurs with 5 to 10 mg/day CPA, and azoospermia occurs only occasionally. Conversely, a combination of testosterone enanthate injections with 12.5 to 100 mg/day CPA results in azoospermia in most men.

Yoshio Okamoto (岡本佳男; born 10 January 1941) is a Japanese chemist, who was awarded the 2019 Japan Prize for his groundbreaking work in asymmetric polymerization and its practical applications in drug discovery. Okamoto was the first to prove that synthetic polymer conformations could be controllable, publishing work on asymmetric polymerization from 1979 onwards. This led to the development by Okamoto and others of helical polymers for use in high performance liquid chromatography columns (HPLC), enabling easy separation of chiral drug molecules.
=== Fragment based (group contribution) === Analogously, the "partition coefficient"—a measurement of differential solubility and itself a component of QSAR predictions—can be predicted either by atomic methods (known as "XLogP" or "ALogP") or by chemical fragment methods (known as "CLogP" and other variations). It has been shown that the logP of compound can be determined by the sum of its fragments; fragment-based methods are generally accepted as better predictors than atomic-based methods. Fragmentary values have been determined statistically, based on empirical data for known logP values. This method gives mixed results and is generally not trusted to have accuracy of more than ±0.1 units. Group or fragment-based QSAR is also known as GQSAR. GQSAR allows flexibility to study various molecular fragments of interest in relation to the variation in biological response. The molecular fragments could be substituents at various substitution sites in congeneric set of molecules or could be on the basis of pre-defined chemical rules in case of non-congeneric sets. GQSAR also considers cross-terms fragment descriptors, which could be helpful in identification of key fragment interactions in determining variation of activity. Lead discovery using fragnomics is an emerging paradigm. In this context FB-QSAR proves to be a promising strategy for fragment library design and in fragment-to-lead identification endeavours. An advanced approach on fragment or group-based QSAR based on the concept of pharmacophore-similarity is developed.

The version carrying the affinity complex bears a maleimide group that binds serum albumin, which extends its circulation time to several days. The version without it lacks this group and clears within roughly half an hour. The two are chemically related but behave very differently once in the body.
In common usage the name without the affinity complex is often equated with MOD GRF 1-29, a fragment carrying four stabilizing substitutions. Strictly speaking, the term originally referred to the albumin-binding version. The overlap in naming causes frequent ambiguity in both informal and technical writing.
Reversed-phase chromatography separates the peptide from related impurities and yields a purity estimate. Mass spectrometry confirms the molecular mass and detects modifications such as oxidation. Sequence-level checks rely on peptide mapping or amino acid analysis when stronger confirmation is needed.
Cool, dark, and dry conditions are standard, with storage at minus twenty degrees Celsius or below. Desiccant and sealed vials limit moisture uptake. Repeated warming and cooling of the container is generally avoided.
